Complement activation correlates with graft damage in baboon-to-human liver xenotransplantation.

چکیده
COMPLEMENT activation has a critical role in the rejection of vascularized organs transplanted between related and widely separated species. I .2 In June 1992 and in January 1993 we undertook two baboon-tohuman liver xenotransplantations. The serum levels ofC3. C4, total hemolytic complement activity (CH 11Xl)' and circulating immune complexes (IC) were monitored. The objective was to see if there was an association of antibody and complement activation with the rejection of these liver xenografts.

Detection of infectious baboon cytomegalovirus after baboon-to-human liver xenotransplantation.
Xenotransplantation is considered to be a solution for the human donor shortage. However, there is a potential risk of transmitting animal infections from the transplanted organ. The known transmissibility and clinical significance of human cytomegalovirus (HCMV) infection after allotransplantation led us to evaluate whether baboon cytomegalovirus (BCMV) transmission could occur after a baboon-...

Aspartate transaminase (AST) and alanine transaminase (ALT) are measured following liver transplantation as indicators of hepatocellular injury. During a series of orthotopic liver allo-and xenotransplants, we observed that there was an increase in AST in all cases. The anticipated concomitant rise in ALT did not occur when a wild-type (WT) pig was the source of the liver graft, but did occur w...
متن کاملHeteroagglutinins and their significance in baboon hepatic xenotransplantation.
The role of preformed xenoreactive antibodies in xenograft recipients is unknown. Humans and baboons possess red cell agglutinating activity associated with isohemagglutinins and heteroagglutinins (HA). We examined the role of HA in two patients who received ABO-identical baboon livers. Human antibaboon HA were assessed by correlating serial titers with studies for rejection. Serial direct anti...
متن کاملClinical xenotransplantation.
Two baboon liver xenografts transplanted to patients with B virus hepatitis supported life for 70 and 26 days but did not function optimally despite minimum or no histopathologic findings of overt humoral or cellular rejection in serial biopsies. However, there was evidence of complement activation in both cases, which in retrospect was thought to explain the unsatisfactory outcome. Strategies ...
متن کاملControl of IBMIR in Neonatal Porcine Islet Xenotransplantation in Baboons
The instant blood-mediated inflammatory reaction (IBMIR) is a major obstacle to the engraftment of intraportal pig islet xenografts in primates. Higher expression of the galactose-α1,3-galactose (αGal) xenoantigen on neonatal islet cell clusters (NICC) than on adult pig islets may provoke a stronger reaction, but this has not been tested in the baboon model. Here, we report that WT pig NICC xen...
